dc.description.abstract | Pavuková, S.:Vinca minor L. alkaloids and their biological activity II. Diploma thesis, Charles University, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany, Hradec Králové 2020. Vinca minor L. is a species of species of flowering plant, native mainly to central and southern Europe, which containst more than 50 indole alkaloids. During screening of potential plant inhibitors against human acetylcholinesterase (hAChE) and butyrylcholinesterase (HBChE) at our department, an alkaloidal extract from dried aerial parts of Vinca minor demonstrated strong and selective hBChE inhibitory activity with an IC50 value of 13.60 ± 0.83 μg/mL, however, against hAChE was inactive (IC50 value >100 μg/mL). The fraction VM 323 - 327 (4,72 g) was separated by column chromatography on silica gel again with stepwise elution by using chloroform and ethanol and overall 7 joined fractions were obtained.Subsequently, repeated preparative TLC on silica gel led to isolation of three compounds; the newly isolated substance SP-1, (-)-picrinine (SP-2) and deacetylakuammiline (SP-3). Diplomová práca, Univerzita Karlova, Farmaceutická fakulta v Hradci Králové, Katedra farmaceutickej botaniky, Hradec Králové 2021. Vinca minor L. je druh kvitnúceho polokru, pôvodom hlavne zo strednej a južnej Európy, ktorý obsahuje viac než 50 indolových alkaloidov. Pri screeningu potenciálnych rastlinných inhibítorov namierených proti ľudskej AChE a BuChE, preukázal alkaloidný extrakt zo sušených nadzemných častí Vinca minor silnú a selektívnu inhibičnú aktivitu namierenú voči BuChE s hodnotou IC50 13,60 ± 0,83 µg/ml avšak proti AChE bol neaktívny (hodnota IC50 >100 µg/ml). Vybraná frakcia VM 323 - 327 (4,72 g) bola rozdelená pomocou stĺpcovej chromatografie na silikagelu stupňovitou elúciou za použitia chloroformu a etanolu a celkovo sa získalo 7 spojených frakcií. Opakovaná preparatívna TLC na silikagelu viedla k izolácii troch zlúčenín; novo vyizolovanej látky SP-1, (-)-pikrininu (SP-2) a deacetylakuammilinu (SP-3). K objasneniu ich štruktúry bola použitá hmotnostná spektrometria (ESI), NMR a optická otáčavosť. Izolované alkaloidy boli podrobené testom inhibičných aktivít voči AChE, BuChE, POP a GSK-3β, ezýmom zohrávajúcim dôležitú úlohu v patofyziológii AD.
