CART peptide, PC12, c-Jun, JNK, PrRP, GPR10, NPFF2, Y5, food intake, lipidizationDisertační práce je zaměřena na studium anorexigenních neuropeptidů, peptidu kokainem a amfetaminem regulovaného transkriptu (CART) a peptidu uvolňujícího prolaktin (PrRP), které snižují příjem potravy a tělesnou hmotnost. Peptid CART je anorexigenní neuropeptid, u kterého přes všechny snahy nebyl nalezen jeho receptor. Peptid CART se specificky váže se na buněčnou linii PC12. Buňky PC12 diferencované v neuronový fenotyp vykazují vyšší počet vazebných míst oproti nediferencovaným buňkám. Buňky PC12 diferencované dexametasonem na chromafinní buňky vykazují vysokou nespecifickou vazbu a tím velmi nízký počet vazebných míst. Pro objasnění důležitosti disulfidových můstků ve struktuře peptidu CART byly syntetizovány analogy s jedním či dvěma disulfidovými můstky. Biologická aktivita byla u analogu se dvěma disulfidovými můstky v pozici 74-94 a 88-101. Při zkoumání buněčné signalizace u buněk PC12 peptid CART významně zvýšil aktivaci c-Jun a fosforylaci JNK, která předchází dráze c-Jun. PrRP patří do rodiny RF-amidových peptidů a byl u něj potvrzen anorexigenní účinek. PrRP se váže na svůj receptor GPR10 a vykazuje vysokou afinitu i k receptoru pro neuropeptid FF (NPFF2). This work focuses on anorexigenic neuropeptides, cocaine- and amphetamine-regulated transcript (CART) and prolactin-releasing peptide (PrRP), which decrease food intake and body weight. CART peptide is an anorexigenic neuropeptide and, despite many efforts, its receptor has not yet been identified. We found CART peptide specific binding sites in pheochromocytoma PC12 cells. Cells differentiated to neurons increased significantly the number of binding sites. On the other hand, after differentiation to chromaffin cells the number of binding sites was so low that it was impossible to determine their density. To clarify the importance of each of the three disulfide bridges in the CART molecule, analogs with one or two disulfide bridges were synthetized. The biological activity was maintained in analog with two disulfide bridges in positions 74-94 and 88-101. Moreover, we demonstrated the stimulation of JNK and subsequently c-Jun activation in PC12 cells. Neuropeptide PrRP belongs to the RF-amide peptide family and has anorexigenic properties. PrPR has a high affinity to GPR10 and neuropeptide FF (NPFF2) receptor.
