dc.description.abstract | Možnosti léčby karcinomů slinných žláz (SGC), obzvláště pokročilých, jsou omezené. Velký pokrok a změnu v léčbě zhoubných nádorů přinesla imunoterapie, zejména terapie inhibitory imunitních kontrolních bodů - immune check-point inhibitors (ICI). Efekt a reakce na imunoterapii pomocí ICI je z velké části řízena charakteristikami imunitních buněk v nádorové tkáni a jak se ukazuje také v peritumorální tkáni. Provedli jsme imunohistochemickou analýzu exprese imunitních kontrolních bodů proteinu PD-1 a ligandu PD-L1 na povrchu nádorových buněk i nádor infiltrujících imunitních buněk (tumor infiltrating immune cells TIIC) ve vzorcích salivárních karcinomů odděleně v jejich středu a na jejich periferii. Kromě výše uvedeného stále větší množství důkazů naznačuje, že odolnost vůči ICI terapii je modulována interakcí Fas receporu (CD95) a Fas ligandu (FasL, CD178) mezi nádorovými buňkami a imunitními buňkami. Rozhodli jsme se proto prozkoumat expresi a interakci Fas-FasL mezi nádorovými buňkami a imunitními buňkami infiltrujícími nádor v jeho středu a v peritumorální oblasti vzorků salivárních karcinomů. Diferenciální hodnocení centra nádoru a periferie nádoru napříč různými histologickými podtypy SGC odhalily roli periferních TIIC a nádorových buněk v pochopení faktorů, které určují závažnost onemocnění.... | cs_CZ |
dc.description.abstract | Treatment options for salivary gland carcinomas (SGC), especially advanced ones, are limited. Immunotherapy, particularly therapy with immune checkpoint inhibitors (ICI), has brought significant progress and change in the treatment of malignant tumors. The effect and response to immunotherapy using ICI are largely driven by the characteristics of immune cells in the tumor tissue and, as it turns out, also in the peritumoral tissue. We conducted an immunohistochemical analysis of the expression of the immune checkpoint protein PD-1 and its ligand PD-L1 on the surface of tumor cells as well as tumor-infiltrating immune cells (TIIC) in samples of salivary carcinomas, separately in their centre and at their periphery. In addition to the above, an increasing amount of evidence suggests that resistance to ICI therapy is modulated by the interaction of the Fas receptor (CD95) and Fas ligand (FasL, CD178) between tumor cells and immune cells.
