Student: Adéla Roztočilová Název diplomové práce: Polymerní nanočástice jako platforma pro dodání ve vodě špatně rozpustných léčiv Polymerní nanočástice mohou být využívány díky jejich výhodným vlastnostem jako nosiče léčiv. Jednou z výhod polymerních nanočástic je např. zvýšení rozpustnosti léčiv ve vodě. Mají velký potenciál v terapii zánětlivých onemocnění jako je Crohnova choroba či v posunu farmakoterapie rakoviny. Hlavním cílem této diplomové práce bylo připravit polymerní nanočástice s enkapsulovaným kurkuminem jako modelové účinné látky. Pro přípravu nanočástic byly použity čtyři typy kopolymeru kyseliny poly(mléčné-ko-glykolové) (PLGA). Byla využita metoda nanoprecipitace a emulzní odpařovací metoda. Hodnocenými parametry byla enkapsulační efektivita, drug loading, velikost částic a polydisperzita. Během experimentu se měnila jak vodná fáze, kde se pracovalo se dvěma různými surfaktanty, tak organická fáze. V rámci experimentální části byla využita rovněž metoda disoluce, kde se porovnávaly dva typy kopolymeru PLGA a v průběhu disoluce byly odebírány vzorky v předem stanovených časech za stanovení celkového uvolněného množství kurkuminu. One of the advantages of polymeric nanoparticles is, for example, to increase the solubility of drugs in water. They have great potential in the therapy of inflammatory diseases such as Crohn's disease or in shifting cancer pharmacotherapy. The main objective of this thesis was to prepare polymeric nanoparticles with encapsulated curcumin as model active ingredient. Four types of poly(lactic-co-glycolic acid) (PLGA) copolymer were used to prepare the nanoparticles. Nanoprecipitation and emulsion evaporation methods were employed. The parameters evaluated were encapsulation efficiency, drug loading, particle size and polydispersity.
