Leukémie je charakterizována akumulací nezralých, abnormálních krevních buněk v krvi a kostní dřeni, které kompetují s normálními hematopoetickými buňkami. Tento stav vede k selhání normální hematopoézy, projevujícímu se anemií, infekcemi a krvácením, leukemické buňky navíc mohou invadovat do ostatních tkání a orgánů. Potlačení normální krvetvorby souvisí s klinickými symptomy onemocnění. Na základě klinických projevů se leukémie dělí na akutní (AL) a chronické a na základě typů buněk, ze kterých pocházejí, do dvou hlavních skupin na Iymfoidní a myeloidní. U dětí tvoří leukémie asi 1/3 všech nádorových onemocně n í. Z toho 80% tvoří akutní lymfoblastická leukémie (ALL), incidence je 3 nové případy /100 000 dětí a adolescentů do 15 věku života za jeden rok. V České republice onemocní ročně asi 65-70 dětí. Dalších 15-20% tvoří akutní myeloidní leukémie a zbylé případy jsou chronické myeloidní leukémie a ostatní typy příbuzné leukémie. Nejčastěji je toto onemocnění diagnostikováno mezi 2. a 6. rokem života. Molekulární analýzy běžných genetických abnormalit v leukemických buňkách přispěly k porozumění patogeneze a ke zlepšení prognózy ALL u dět í (1 ,2 ). Chromosomální přestavby u tohoto onemoc nění často přítomné způsobují aberantní expresi protoonkogenů a tvoří fúzní geny, které kódují konstitutivně aktivní...
