dc.description.abstract | Úvod: Výskyt diseminovaných nádorových buněk (DTCs) a cirkulujících nádorových buněk (CTCs) je spojen se špatnou prognózou a zvýšeným výskytem úmrtí v důsledku onemocnění u pacientek s nemetastatickým karcinomem prsu. Cílem dizertační práce bylo stanovit DTCs v kostní dřeni a CTCs v periferní krvi u pacientek s primárním karcinomem prsu, zhodnotit korelaci výskytu DTCs/CTCs s přítomností dalších prognostických markerů a monitorovat výskyt DTCs/CTCs v průběhu léčby. Metody: Do studie bylo zařazeno 50 pacientek s primárním karcinomem prsu. K analýze CTCs z periferní krve byla použita imunomagnetická separace a k jejich detekci byl použit diagnostický kit AdnaTest Breast Cancer™ (AdnaGen GmbH, Langenhagen, Německo). Detekce DTCs v kostní dřeni probíhala pomocí imunocytochemické analýzy s pomocí pancytokeratin protilátky konjugované s FITC (monoklonální anti-cytokeratin protilátka F3418, Sigma Aldrich, USA). Výsledky: DTCs jsme stanovili u 30% (15/50) a CTCs u 22% (11/50) pacientek. Studie potvrdila, že DTC pozitivita signifikantně korelovala s velikostí primárního nádoru (p- hodnota 0,011) a infiltrací lymfatických uzlin (p-hodnota 0,002). CTC pozitivita nekorelovala ani s velikostí nádoru či infiltrací axilárních uzlin. DTCs byly převážně detekovány u pacientek ER/PR negativních a HER-2 pozitivních.... | cs_CZ |
dc.description.abstract | Introduction: Simultaneous detection of disseminated tumor cells (DTCs) and circulating tumor cells (CTCs) was shown to be associated with an especially poor prognosis and increased incidence of disease-related deaths in non-metastatic breast cancer patients. We analyzed the occurance of DTCs in bone marrow and CTCs in peripheral blood in patients with primary breast cancer, we evaluated the correlation of their presence with other prognostic markers and we investigated the changes in DTCs/CTCs number at different time points during treatment. Materials and methods: Blood of 50 patients with primary breast cancer were used for immunomagnetic separation and detection of circulating tumor cells using the commercial available system the AdnaTest Breast Cancer™ (AdnaGen GmbH, Langenhagen, Germany). Bone marrow aspirates from 50 patients were analyzed for DTCs by immunocytochemistry using the pancytokeratin antibody conjugated with FITC (Monoclonal Anti-Cytokeratin antibody F3418, Sigma Aldrich, USA).
