dc.description.abstract | Cieľom tejto diplomovej práce bolo stanoviť stabilitu doxorubicínu a daunorubicínu, obe látky vo forme hydrochloridu, vo vode, v roztoku pre aplikáciu králikovi (0,9% roztok NaCl) a kultivačnom médiu pomocou HPLC za použitia fluorescenčného detektoru. Doxorubicín a daunorubicín patria do skupiny antracyklínových antibiotík používaných pri liečbe rôznych typoch rakoviny. Bola skúmaná stabilita látok vo vode a 0,9% roztoku NaCl skladovaných v skle a plaste (polypropylén) pri izbovej teplote na svetle a v tme počas 24 hodín, v chladničke (8řC) počas 3 mesiacov, v mrazničke (-80řC) počas 3 mesiacov a opakovane zmrazovaných (-80řC) a rozmrazovaných (izbová teplota), celkovo 3-krát. Stabilita látok v kultivačnom médiu skladovaných v skle a plaste (polypropylén) bola skúmaná pri izbovej teplote na svetle a v tme počas 3 dní. Ďalej boli roztoky látok skladované pri 37řC v termomixéri chránené pred svetlom v plaste (polypropylén) a v inkubátore v skle a plate (polypropylén a polystyrén) a skúmané počas 3 dní. Tiež sa skúmala ich stability pri opakovanom zmrazený (-80řC) a rozmrazení (izbová teplota) neriedených roztokov i roztokov zriedených 90% metanolom. | cs_CZ |
dc.description.abstract | The purpose of this thesis was to determine the stability of doxorubicin and daunorubicin, both in the form of hydrochloride, in water, in a rabbit application solution (0.9% NaCl solution), and culture medium using HPLC with a fluorescence detector. Doxorubicin and daunorubicin belong to the group of anthracycline antibiotics used in the treatment of various types of cancer. The stability of the substances has been investigated in water and 0.9% NaCl stored in glass and plastic (polypropylene) at room temperature in light and dark for 24 hours, in a refrigerator (8řC) for 3 months, in a freezer (-80řC) for 3 months, and repeatedly frozen (-80řC) and thawed (room temperature), totaling 3 times. The stability of the substances in culture medium has been investigated in glass and plastics (polypropylene) at room temperature in light and dark for 3 days Additionally, stock solutions were stored at 37řC in thermomixers protected from light in plastics (polypropylene) and in an incubator in glass and plastics (polypropylene and polystyrene) and examined for 3 days. They were also examined for their stability at repeated frozen (-80řC) and thawing (room temperature) of undiluted solutions and solutions diluted with 90% methanol. | en_US |
