Akutní hypoxie nebo ischémie patří v neurologii k etiologicky nejvýznamnějším jednotkám vedoucím k náhlé mozkové dysfunkci. V časné fázi je jejím podkladem membránová depolarizace vedoucí k nadměrnému přesunu iontů a vody. Buněčný edém vede ke změnše n í velikosti ECP a tedy i zvýšení koncetrace látek v něm obsažených, včetně toxických metabolitů. Délka trvání patologického podnětu je určuj ícím faktorem pro vznik nevratného buněčné poškození formou apopotózy nebo nekrózy. Změny difúzních parametrů způsobené hypoxickým podnětem byly e x p e rim entálně studovány dosud pouze na modelu globální anoxie navozené srdeční zástavou. Na tomto modelu byly zjištěny dramatické změny ve velikosti a tortuozitě ECP mozkové k ůry , které se rozvinuly během několika minut po zástavě mozkové perfúze. Rozhodli jsme se proto zkoumat změny difúzních parametrů ECP mozkové kůry i na modelech, kde délka trvání patologického podnětu byla časově omezená. Jednalo se o model tranzientní hypoxie a tranzientní hypoxie/ischemie. Získaná data jsme korelovali se změnam i v extracelulární koncetraci laktátu, glukózy a glutamátu. Během tranzientní hypoxie/ischémie jsme zaznamenali postupný rozvoj změn difúzních parametrů ECP po celou dobu trvání podnětu.
