Stanovení cytokinů v supernatantu homogenátu jater u potkanů s jaterní steatózou

Název diplomové práce: Stanovení cytokinů v supernatantu homogenátu jater u potkanů s jaterní steatózou Zaměřením této práce bylo stanovení cytokinů, a to IL-6, TNF-alfa a leptinu v supernatantu homogenátu jater u potkanů s jaterní steatózou. Tato vyšetření byla vykonána v rámci rozsáhlé experimentální práce orientované na schopnost jater regenerovat po indukci steatózy. Stanovení byla provedena pomocí enzymové imunoanalýzy na imunosorbentech. Výsledky byly porovnávány s koncentracemi cytokinů kontrolních skupin potkanů kmene Wistar a kmene PHHP, kteří byli krmeni standardní laboratorní dietou. U kontrolní skupiny potkanů kmene Wistar byly koncentrace IL-6, leptinu i TNF-alfa v supernatantu jaterní tkáně vyšší než u kontrolní skupiny potkanů kmene PHHP. Příjem steatogenních diet vedl spíše k poklesu IL-6 u všech souborů potkanů. U pokusné skupiny kmene Wistar byly koncentrace leptinu signifikantně nižší oproti kontrolám téhož kmene. U kmene PHHP podávání orotové kyseliny vedlo ke statisticky významnému zvýšení koncentrace leptinu. Title of diploma thesis: Cytokine estimation in liver homogenate supernatant in rats with liver steatosis The aim of this work was the determination of cytokines - interleukin 6, tumor necrosis factor-alpha and leptin levels in liver homogenate supernatant in rats with hepatic steatosis. These tests were performed within extensive experimental work intented on the ability of liver regeneration after induction of steatosis. The determination was performed using enzyme-linked immunosorbent assay. The results were compared with cytokine concentrations of Wistar and PHHP control groups, who were fed a standard laboratory diet. The concentrations of IL-6, leptin and TNF-alpha in liver tissue supernatant were higher in the control group of Wistar rats than in PHHP rats control group. Receiving steatosis diets led to a decrease in IL-6 for all rat files. Leptin concentrations were significant lower in Wistar rats than Wistar rats controls were.
